ABSTRACT

Matrix-assisted laser desorption/ionization mass spectrometry imaging (MALDI-MSI) is a new imaging technique with label-free, rapid, and high throughput features. It has bloomed in the analysis on the spatial distribution of biomolecules such as drugs, metabolites, peptides and proteins on the tissue surface in virtue of providing high data throughput from non-targeted full analysis and high accuracy from targeted analysis. The acquisition of MSI signal response with high sensitivity, high spatial resolution, and good stability is directly depended on the appropriate sample preparation approaches, and flexible and various data processing tools will help the non-target data mining to meet the demands of visualization, spatial distribution and multiple index applications so as to reveal the scientific rules beneath the data. This review briefly summarizes the key advances in MALDI-MSI from aspects of sample preparation procedures, data processing and visualization. It also illustrates the characteristics, difficulties and probable solutions derived from these key techniques.

ABSTRACT

Radiation-induced intestinal injury(RIII), a common complication of radiotherapy for pelvic malignancies, affects the quality of life and the radiotherapy efficacy for cancer. Currently, the main clinical approaches for the prevention and treatment of RIII include drug therapy, hyperbaric oxygen therapy, and surgical treatment. Among these methods, drug therapy is cost-effective. Traditional Chinese medicine(TCM) containing a variety of active components demonstrates mild side effects and good efficacy in preventing and treating RIII. Studies have proven that TCM active components, such as flavonoids, terpenoids, phenylpropanoids, and alkaloids, can protect the intestine against RIII by inhibiting oxidative stress, regulating the expression of inflammatory cytokines, modulating the mitochondrial apoptosis pathway, adjusting intestinal flora, and suppressing cell apoptosis. These mechanisms can help alleviate the symptoms of RIII. The paper aims to provide a theoretical reference for the discovery of new drugs for the prevention and treatment of RIII by reviewing the literature on TCM active components in the last 10 years.

ABSTRACT

Alfalfa (Medicago sativa) is an important food and feed crop which rich in mineral sources. The WUSCHEL-related homeobox (WOX) gene family plays important roles in plant development and identification of putative gene families, their structure, and potential functions is a primary step for not only understanding the genetic mechanisms behind various biological process but also for genetic improvement. A variety of computational tools, including MAFFT, HMMER, hidden Markov models, Pfam, SMART, MEGA, ProtTest, BLASTn, and BRAD, among others, were used. We identified 34 MsWOX genes based on a systematic analysis of the alfalfa plant genome spread in eight chromosomes. This is an expansion of the gene family which we attribute to observed chromosomal duplications. Sequence alignment analysis revealed 61 conserved proteins containing a homeodomain. Phylogenetic study sung reveal five evolutionary clades with 15 motif distributions. Gene structure analysis reveals various exon, intron, and untranslated structures which are consistent in genes from similar clades. Functional analysis prediction of promoter regions reveals various transcription binding sites containing key growth, development, and stress-responsive transcription factor families such as MYB, ERF, AP2, and NAC which are spread across the genes. Most of the genes are predicted to be in the nucleus. Also, there are duplication events in some genes which explain the expansion of the family. The present research provides a clue on the potential roles of MsWOX family genes that will be useful for further understanding their functional roles in alfalfa plants.

ABSTRACT

Objective@#To investigate clinicopathological features and prognostic factors of gastric neuroendocrine tumors (G-NEN).@*Methods@#Clinical and pathological data of patients with G-NEN diagnosed by pathological examination in Chinese PLA General Hospital from January 2000 to June 2018 were retrospectively analyzed in this case-control study. Patients with complicated visceral lesions, other visceral primary tumors, mental disorders and incomplete clinicopathological data were excluded. Finally, 240 hospitalized patients who met the inclusion criteria were enrolled. Physical examination information, tumor characteristics and pathological characteristics of patients were summarized. The Cox regression models were used to analyze the risk factors affecting G-NEN and the survival conditions were described by Kaplan-Meier survival curves and log-rank test.@*Results@#In 240 patients with G-NEN, the mean age was (60.3±10.1) years; 181 were male (75.4%) and 59 females (24.6%); mean tumor diameter was (4.2±2.8) cm; 51 cases (21.2%) were neuroendocrine tumor (NET), 139 cases (57.9%) neuroendocrine carcinoma (NEC), 50 cases (20.8%) mixed neuroendocrine carcinoma (MANEC); 28 cases (11.7%) were G1 low grades, 34 cases (14.2%) G2 medium grades, and 178 cases (74.2%) G3 high grades; tumor infiltration depth T1 to T4 were 44 cases (18.3%), 27 cases (11.2%), 60 cases (25.0%) and 109 cases (45.4%) respectively; 163 cases (67.9%) developed lymphatic metastasis and 46 patients (19.2%) distant metastasis; tumor stage from stage I to stage IV were 55 cases (22.9%), 42 cases (17.5%), 94 cases (39.2%) and 53 cases (22.1%) respectively. Of the 240 G-NEN patients, 223 cases (92.9%) were followed up. The median survival time of the patients was 39.2 (95% CI: 29.1 to 47.5) months. Univariate survival analysis showed that age ≥ 60 years, tumor diameter ≥ 4.2 cm, tumor grade G3, lymphatic metastasis, distant metastasis, and tumor stage III-IV were risk factors for G-NEN patients. Multivariate survival analysis revealed that lymphatic metastasis (HR=1.783, 95%CI: 1.007-3.155, P=0.047) and distant metastasis (HR=2.288, 95% CI: 1.307-4.008, P=0.004) were independent risk factors of the prognosis. Further analysis of the G3 subgroup of G-NEN showed that the 5-year survival rate of NET-G3 was 76.19%, which was significantly higher than that of NEC-G3 and MANEC-G3 (15.60% and 24.73%, P=0.012).@*Conclusions@#Most G-NEN patients are in advanced stage at diagnosis. Lymphatic metastasis and distant metastasis indicate poor prognosis. The prognosis of high proliferation NET-G3 patients is better as compared to those of NEC-G3 and MANEC-G3. This classification is worth further attention.

ABSTRACT

Purpose To investigate the expression characteristics and significance of P-selectin and HPA in benign and malignant hydatidform mole tissues. Methods By using immunohistochemical technique SP and RT-PCR, the expression of Pselectin and HPA was detected in 40 cases of benign hydatidiform mole tissues, and 12 malignant hydatidiform mole tissues.Meanwhile, 40 cases of normal placental villi tissues were observed. Results The protein positive rate and protein relative quantity expression and the mRNA relative quantity expression of P-selectin and HPA were gradually increasing in normal placenta villi tissues, benign hydatidiform mole tissues, and malignant hydatidiform mole tissues. The differences between groups were statistically significant (all P<0.05). The expression of P-selectin and HPA showed positive correlation (all P<0.05) in benign and malignant hydatidform mole tissues. Conclusion The over-expression of P-selectin and HPA promotes the occurrence and development of hydatidform mole. The detection of the expression levels of P-selectin and HPA is of great significance for the differential diagnosis and prognosis assessment of benign and malignant hydatidform mole. It could become a biological predictor of malignant development of hydatidform mole.

ABSTRACT

Objective To establish a species identification system based on DNA genetic markers for plant evidence. Methods Two hundred common plants in Shanghai were collected and identified by mor-phological characteristics. The primers of gene segments rbcL, matK, and ITS were designed and amplified. The PCR amplicon was detected by agarose gel electrophoresis. After the sequencing, the universality and the identification capacity of the three markers were evaluated. Results The success rate of amplifi-cation was in order of rbcL (99.5%) >matK (92.5%) > ITS (86.0%). The identification capacity of the combination of rbcL and matK was better than that of rbcL or matK, by which most plant species could be identified to the genus or higher. ITS was not suitable to be a unique marker because of its unstable result, but it still could be a powerful supplement. The identification capacity of the combination of rbcL, matK and ITS was higher than that of rbcL and matK, by which most plant species could be identified to the genus or lower. Conclusion The identification system with the combination of rbcL, matK and ITS as markers has excellent universality for plant evidence, which can distinguish most plant species to the genus or lower.

ABSTRACT

AIM:To investigate the effect of microRNA(miR)-381-3p on neuronal injury in the hippocampus of depressive rats and the possible regulatory mechanism.METHODS:The rat model of depression was established by sub-cutaneous injection of corticosterone and the model rats received fluoxetine treatment. The body weight, open field test and biochemical indexes were measured for judging the therapeutic effect of fluoxetine. The expression of miR-381-3p,brain-derived neurotrophic factor(BNDF),Bcl-2 and Bax in the hippocampus was determined. The cultured neonatal rat hipp-ocampal neurons were pre-transfected with miR-381-3p inhibitor and then incubated with corticosterone. The cell viability, and the expression of miR-381-3p, BNDF, Bcl-2 and Bax were detected. The targeted regulatory role of miR-381-3p in BDNF was verified by luciferase reporter assay.RESULTS:Compared with depression group, fluoxetine increased the body weight, the number of cross-field activities and the content of norepinephrine, inhibited the expression of miR-381-3p and promoted the expression of BNDF in the hippocampus.miR-381-3p silencing reversed the effect of corticosterone, resulting in the increase in the survival rate of hippocampal neurons, upregulation of BDNF and Bcl-2, and downregulation of Bax expression.miR-381-3p targeted the regulation of BNDF expression.CONCLUSION: Silencing miR-381-3p protects rat hippocampal neurons from corticosterone injury, and its mechanism may be related to the upregulation of BNDF expression.

ABSTRACT

With the continuous improvement of endoscopic technology and equipment, minimally invasive treatment of bile duct stones be-comes increasingly diversified. In this paper, the concept of minimally invasive treatment of bile duct stones, surgical indications and contra-indications,surgical methods, and the advantages and disadvantages of surgical methods are introduced. It is shown that the application of laparoscope has laid the foundation for the minimally invasive treatment of bile duct stones, and the combined application of choledochoscope and endoscope has brought new ideas for minimally invasive treatment of bile duct stones. The combination of three endoscopes avoids the disadvantages of traditional open surgery:large trauma and long operation time, and reduces the patient′s pain and complications. In the near future, the combination of three endoscopes still has much room for growth. As long as the indications for several surgeries are mastered and the safety and efficacy are evaluated objectively, the combination of three endoscopes will play the biggest role.

ABSTRACT

Objective To explore the clinical features of type 2 diabetic patients with hypertension , and to analyze the influencing factors of glycemic control. Methods Using stratified cluster random sampling method, 5 communities were selected from urban areas of Huai'an in 2014. Type 2 diabetic patients managed by the communities were surveyed with questionnaire, physical and biochemical examinations. The related information and clinical features were compared between diabetic patients with and without hypertension. Logistic regression analysis was used to analyze the influencing factors of glycemic control. Results The number of well-controlled diabetic patients (HbA1c<7.0%) with and without hypertension (HbA1c<7.0%) were 419 (39.3%) and 480 (52.1%), respectively. Mean values of body mass index (BMI), diabetic duration and serum creatinine in diabetic patients with hypertension were significantly higher than those in diabetic patients without hypertension (P<0.05). The proportions of macrovascular complications and dyslipidemia in diabetic patients with hypertension were significantly higher than diabetic patients without hypertension (P<0.05). Multiple logistic regression analysis showed that high degree of education, high annual family income and high frequency of glucose monitoring were beneficial factors for glycemic control in diabetic patients with hypertension. Older age, hypertension, higher waist to hip ratio (WHR), the elevated triglyceride and serum creatinine were harmful factors for glycemic control. Conclusions The situation of glycemic control in diabetic patients with hypertension in urban areas of Huai'an is not optimistic. Therefore, community managements of risk factors such as central obesity and increased triglyceride in elder diabetic patients should be strengthened.

Objective To examine the impact of stressful training on recruits and relationships with adult attachment, neuroticism and negative coping style. Methods 1 962 recruits were assessed with aduh attachment scale (AAS),impact of event scale-revised (IES-R),Eysenck personality questionnaire (EPQ) and trait coping style questionnaire (TCSQ).Results (1) 67.84% of recruits reported the varying degrees of stress, and the total average of IES-R was 1.25±0.53.The impact of training of only-child recruits (1.28±0.56) was higher than that of none-only-child (1.22±0.51) (P<0.05).The total average of attachment close was 3.73±0.51,attachment depend was 3.24±0.53,and attachment anxiety was 2.40±0.77.(2) Attachment anxiety, neuroticism and negative coping style were all positively correlated with impact of training (r =0.384,0.570,0.543,all P< 0.01).Attachment close and depend were both negatively correlated with impact of training (r=-0.221,-0.251,all P< 0.01).(3) Neuroticism and negative coping style played a completely intermediary role between adult attachment and impact of training.(4) The multiple group analysis showed that the mediation model was adapted to the only-child or none-onlychild recruits. Conclusion Adult attachment can predict impact of training to some extent in the intermediary roles of neuroticism and negative coping style.

ABSTRACT

Objective To explore the relationship between personality and depression, especially to investigate mediating effect of positive mental characters. Methods A sample of 1 933 male recruits was assessed with eysenck personality questionnaire (EPQ),positive mental characters scale for recruits (PMCS-R) and self-rating depression scale (SDS).Results The scores of introversion-extroversion,psychoticism,neuroticism,positive mental characters and depression were 55.26±10.59,42.85±7.25,42.35± 11.67,3.80±0.64,0.45±0.11,respectively.There were correlations among personality, positive mental characters and depression in recruits (P<0.05).Regression analysis indicated that introversion-extroversion,psychoticism and neuroticism predicted depression directly, and it could explain 42.6% of the total variance (F=476.28,P<0.05).Hierarchy regression analysis showed that positive mental characters mediate the relationship between personality (introversion-extroversion,psychoticism,neuroticism) and depression (the mediation effect was 0.22,0.19 and 0.11 respectively,P< 0.05).Conclusion Recruits' personality affects depression not only directly, but also indirectly through positive mental characters.

ABSTRACT

Objective To explore sleep quality and related psychological factors among young army cadres to provide theoretical basis for health interventions. Method A total of 1 100 young army cadres aged 18 to 34 were assessed with Pittsburgh sleep quality index (PSQI), Symptom Checklist-90 (SCL-90) and Center for Epidemiologic Studies Depression Scale by cluster random sampling. Based on CES-C scores, cadres were divided into two groups named depression group and non-depression group;850 sergeants were served as the control group. T test, chi-square test and multiple regressions were used in data analysis. Result Sleep problems were found in 13.5% of the cadres and 12.1% sergeants. General status of sleep quality of cadres was worse than sergeants' and norm's. Exercise less, physical disease and psychological problems were the risk factors. Multiple stepwise regression analysis showed that depression factors of SCL-90 had the most significantly predictive effect on sleep quality (r=0.552, Beta=0.387), followed by somatization (r=0.552, Beta=0.223), two factors could explain 32.7% of PSQI scores variation. Correlation analysis showed that depression is closely related to subjective sleep quality, sleep duration, sleep disorder and daytime dysfunction (all r>0.3). Cadres' depression (11.11 ± 6.62) was significantly lower than norm's (12.30 ± 10.39) and higher than sergeants'(8.11 ± 6.56). PSQI scores of depression group (6.05 ± 3.13) were significantly higher than non-depression group's (3.56 ± 2.69) and norm's (3.23 ± 3.12). Conclusion Depression is a major correlation factor of the sleep quality. Regular exercise, improvement of the physical and psychological health quality, and reducing depression are effective ways to improve sleep quality.

ABSTRACT

The cAMP-responsive element binding protein (CREB)-regulated transcription coactivator, CRTC (also known as transducer of regulated CREB, TORC), is identified as a potent modulator of cAMP response element (CRE)-driven gene transcription. The CRTC family consists of three members (CRTC1-3), among which the CRTC1 shows the highest expression in the brain. Several studies have demonstrated that the CRTC1 plays critical roles in neuronal dendritic growth, long-term synaptic plasticity, memory consolidation and reconsolidation etc., whereas dysfunction of CRTC1 is mainly involved in neurodegenerative disorders. In light of these findings, we aim to review recent research reports that indicate the CRTC1 dysfunction and its underlying mechanisms in the neurodegenerative disorders.
